Aim of this protocol is to describe construction of large and diverse recombinant antibody libraries to isolate specific lead antibodies using yeast display. To isolate specific lead antibodies first and most important step is to construct large and diverse antibody libraries so that panel of antibodies can be selected from them. For isolation of panel of antibodies; diverse and large antibody libraries have been constructed using recombinant DNA technology, genetic and protein engineering technologies.  For selection of panel of antibodies, yeast display one of the best approaches that have been used successfully. In this protocol, construction of large and diverse antibody libraries and antibodies displayed in yeast display format have been described to select and isolate panel of specific antibodies that have been used successfully.
